چکیده

Background Anthracyclines and Herceptin are well known amongst chemotherapeutic agents to cause cardiotoxicity. Anthracyclines, particularly in combination with, Herceptin are seen in studies to cause cumulative, dose-related, progressive myocardial damage with decrease in left ventricular(LV) ejection fraction (EF) ultimately leading to clinical heart failure, which can be irreversible. There is lack of appropriate guidelines to monitor cardiac function in these patients. We hypothesized that changes in tissue distribution of gadolinium (Gd) and/or myocardial strain identified using (Gd)-enhanced cardiac magnetic resonance (CMR) could help identify early myocardial damage which can lead to systolic dysfunction.

Cardiotoxicity associated with trastuzumab (Herceptin) therapy in the treatment of metastatic breast cancer.

Trastuzumab (Herceptin) is a humanised monoclonal antibody that specifically targets HER2-positive breast cancer cells. Safety data collected from pivotal trials with trastuzumab indicate that this therapy is generally well tolerated. However trials of the combination of trastuzumab plus chemotherapy, and in particular chemotherapy with anthracyclines, have revealed an elevated incidence of car...

Subendocardial late gadolinium enhancement in two patients with anthracycline cardiotoxicity following treatment for Ewing's sarcoma.

Cardiotoxicity is a well-known consequence of anthracycline chemotherapy. We report CMR findings not previously described in two patients with anthracycline cardiotoxicity following treatment for Ewing's sarcoma. Subendocardial enhancement on late gadolinium contrast-enhanced CMR was present in both cases, with histological correlation in one case.

Delayed contrast enhancement cardiac magnetic resonance imaging in trastuzumab induced cardiomyopathy

BACKGROUND Trastuzumab (Herceptin), an antagonist to the human epidermal growth factor 2 (HER2) receptor significantly decreases the rates of breast cancer recurrence and mortality by 50%. Despite therapeutic benefits, the risk of cardiotoxicity with trastuzumab ranges from 10-15% when administered sequentially following anthraycline chemotherapy. Little is known about the utility of cardiac ma...

The Effect of N-Acetylcysteine Administration to Prevent Anthracycline-Induced Cardiotoxicity in Breast Cancer Patients

Introduction: Anthracyclines are one of the classes of chemotherapy drugs that  are widely used to treat many types of cancers including breast cancer. Taking this class of medications has a significant relationship with cardiac dysfunction. N-acetylcysteine has antioxidant properties and may be effective in preventing cardiac dysfunctions. In this study, we investigated the effect of N-acetylc...

Modulation of anthracycline-induced myofibrillar disarray in rat ventricular myocytes by neuregulin-1beta and anti-erbB2: potential mechanism for trastuzumab-induced cardiotoxicity.

BACKGROUND There is an increased incidence of heart failure in patients treated concurrently with anthracyclines and the chemotherapeutic anti-erbB2 agent trastuzumab (Herceptin). On the basis of our previous studies with recombinant neuregulin-1beta (NRG-1beta), a ligand for the erbB2 receptor tyrosine kinase, we hypothesized that activation of erbB2 by anti-erbB2 versus NRG-1 would cause diff...
